Helicase targeting An appropriate approach to fight COVID-19 infection

چکیده
Statement of Problem Respiratory infections are one of the most common infections in the world and take many victims every year. Severe Acute Respiratory Syndrome Coronavirus 2 (SARS-CoV-2) which was identified in Wuhan China about two years ago is one of the causes of a type of respiratory infection called COVID-19. So far more than 280 million people have been infected with Covid-19 worldwide of which more than 5 million have died. Therefore finding solutions to prevent and treat this disease is necessary. So far many natural products have been identified to fight infections caused by a variety of coronaviruses. These compounds can target virus components and proteins such as spike (S) helicase nucleoprotein (N) and 3A or host cell proteins including Angiotensin-converting enzyme 2 (ACE2) as specific receptors for SARS-CoV-2 and Transmembrane serine protease 2 (TMPRSS2) as a facilitator of virus entry into the host. Research Purpose The present study aimed to review the natural compounds affecting respiratory infection caused by the SARS-CoV-2 virus and to select the most appropriate compound. Results and Conclusion The findings of this study indicate that the coding regions of components such as S and N are very prone to mutations and various mutations have occurred in them thus far particularly in the newest variant Omicron. Therefore if a major change in the structure of proteins occurs due to the increase in mutations in these regions it is obvious that the products against them will no longer have the desired effect. On the other hand natural products affecting these proteins also harm similar proteins or their receptors in the host cell which can disrupt the normal functioning of the host cells. Therefore it is more desirable to use compounds that have fewer side effects on the host cell and affect the virus in a more targeted way. Mutations occur infrequently in the helicase coding region that is part of ORF1b. In addition no report has been published on the anti-viral helicase compounds with the effect against host cell helicase. Therefore the use of viral helicase inhibitors can be a good way to fight SARS-CoV-2 infection. The most important of these compounds are bananins which are synthesized by the reaction of the aldehyde form of vitamin B6 with phloroglucinol and inhibit the continuation of virus replication by inhibiting ATPase and unwinding activities of helicase.کليدواژه ها
